The High Throughput Cell-based Screening Facility (HTSF) of the Institute of Genetics and Molecular and Cellular Biology (IGBMC) provides to researchers an infrastructure for high throughput cell imaging and a global solution for large genetic gain/loss of function or chemical compound screenings.
Cell-based assays and image-based high content analyses are designed, developed, miniaturized, optimized by the members of the facility together with the research teams. Our gene-oriented technologies rely on RNA interference and the CRISPR/Cas9 technology.
The primary platform mission is to support basic and translational medical research on various topics studied by the four main research departments of the institute and in particular cancer and rare genetic diseases. Associated to the Pôle de Competitivité Alsace-Biovalley, Conectus-Alsace and the Canceropôle Grand Est, the platform is open to academic research outside the institute and also to private companies.
Understanding the molecular basis of biological processes or cellular phenotypes using gene knockdown is achieved using mammalian cell transfection of synthetic small interfering siRNA molecules and single cell imaging using dedicated automated devices enabling throughputs for genome-wide siRNA-based screening projects. Cell models and assays are usually provided by research groups but can also be developed by the facility itself (fluorescent reporter cell engineering, CRISPR/Cas9 vector production, antibody production, …).
Identifying lead compounds and drugs towards a biological process or a cellular phenotype is achieved using for example the Prestwick and the SelleckChem FDA-approved chemical libraries.
High Content Analysis and Screening is performed using Thermofisher Scientific Cellomics ArrayScan XTi and CellInsight CX5/7 readers.
